In collaboration with Mike Schutkowski lab (Halle, Germany) we have developed a new continuous fluorescent sirtuin activity assay using the native myristoyl residue at the lysine side chain of a substrate. Excellent kinetic parameters of new substrates allowed to reliably determine inhibition constants for enzyme concentrations as low as 50 pM using different microtiter plate formats.
