Our manuscript “Functional Divergence and Structural Changes of Class IV Histone Deacetylases (HDACs) Across the Tree of Life” has been accepted for publication. In this study, we combined phylogenetics, structural biology, enzymology, and mass spectrometry to uncover two deeply conserved HDAC11 lineages with distinct localization, structure, and substrate preferences. The work provides a revised evolutionary framework for class IV HDACs and new insight into the functional diversification of this ancient enzyme family.
